Debería probar con el operador de igualdad, no de asignación:

Código Delphi [-]
if RadioButton1.Checked = true then
  {...}

Pero, aún se puede resumir, en este caso:

Código Delphi [-]
if RadioButton1.Checked then
  {...}

O bien:

Código Delphi [-]
if not RadioButton1.Checked then
  {...}
